The LumiPure genomic DNA from AnySample Kit is designed to isolate genomic DNA from a wide range of biological samples using spin columns. The kit includes all the necessary components. First, cell lysis is performed using Lysis Solution and Proteinase K. The lysate is mixed with Sorption Solution that allows selective binding of DNA to the membrane in the column. In the following step, a series of washings of bound DNA are performed. Special formulations of Washing Solutions allow impurities to be completely removed from the column membrane. Finally, the genomic DNA is eluted with Elution Buffer or, if necessary, with deionized water. The kit also includes corundum and pestles for homogenization of plant and animal tissues, RNase A for efficient removal of any residual RNA, Red Blood Cell Lysis Buffer (in case of prior separation of leukocytes, which provides increased DNA yield compared to DNA purification from a whole blood sample). Column binding capacity of 20 µg DNA. DNA typical yield: plant and animal tissues – 3–20 µg (depending on the sample); whole blood (100 µL) – 1–3 µg; leukocytes (isolated from 1 mL of whole blood) – 4-5 µg; buccal swab – 0.5-2 µg; cultured cells (1 Î 10⁶) – 3-6 µg; gram-negative bacteria (1 Î 10⁸) – 5–10 µg. DNA quality of 30–50 kb in size, OD 260/280 of 1.7–1.8. The isolated DNA is suitable for PCR, restriction enzyme digestion, Southern blotting, preparing samples for Sanger sequencing and NGS, etc.
